Method Description:

Sample is extracted with a buffered salt solution, followed by centrifugation of filtration. Sample is added to antibody coated wells and any unbound residue is washed away. A second enzyme-labeled antibody is added. Color develops and a stop reagent is added. A microwell reader is used to determine optical densities. Sample optical densities are plotted against control optical densities to determine exact egg residue concentration.

The egg allergen test is a sandwich enzyme-linked immunoassay (ELISA) that measures egg protein.

Concentrated food additives, colors, and flavors may cause interferences on ELISA test methods.

Hydrolyzed and fermented proteins may not be detected using ELISA methods for allergen testing. Due to the breakdown of the proteins into small peptides or amino acids, they may become undetectable by this assay, but still could be allergenic and cause an allergic reaction.
