Method Description:

This method detects and quantitates the common phenolic antioxidants: propyl gallate (PG), 2,4,5-trihydroxybutyrophenone (THBP), tert-butylhydroquinone (TBHQ), nordihydroguaiaraetic acid (NDGA), 2-and 3-tert-butyl-4-hydroxyanisole (BHA), 2,6-di-tert-butyl-4-hydroxymethylphenol (Ionox-100), and 2,6-di-tert-butyl-4-hydroxytoluene (BHT). Applicable to animal and vegetable fats, oils, lards, shortenings, as well as antioxidant concentrate dissolved in other solvents. Samples are first dissolved in hexane or 1:1 mixture of acetonitrile: 2-propanol. If the sample is soluble in hexane, the antioxidants are partitioned into acetonitrile, which in turn is concentrated and diluted with 2-propanol (isopropanol). If the sample is soluble in a 1:1 mixture of acetonitrile:2-propanol, the sample is diluted. Antioxidants are then separated and quantitated using reverse-phase gradient elution HPLC using ultraviolet detection at 280 nm.
