Method Description:

Samples are incubated with pancreatic a-amylase and amyloglucosidase (AMG) and further treated to remove the non-resistant starch. The resistant starch is then dissolved in KOH, neutralized with acetate buffer, and then is quantitatively hydrolyzed to glucose with AMG. Glucose is measured with glucose oxidase/peroxidase reagent (GOP) on a spectrophotometer. The absorbance of this solution at 510 nm is proportional to the RS content of the sample.


Information Required by Submitter:

Please provide estimates for samples containing high (> 10%) resistant starch. The method must be adjusted slightly for samples with high resistant starch.


Additional Information:

This method measures RS2 (granular) and RS3 (retrograded) starches. RS1 starches are not detected by this method. Applicability of this method to RS4 (chemically-cross linked) type starches must be determined on a case-by-case basis.
